Lokhande was born on December 19, 1984, to her mother Vandana Phadnis Lokhande, a teacher, and her father Shashikant Lokhande, in a Marathi family in Indore. She has two brothers, Sooraj and Arun, as well as a sister, Jyoti. She moved to Mumbai in 2005 to pursue a career in acting after graduating.
Ankita Lokhande announced her relationship with Vicky Jain, a businessman, in 2019. They will marry on December 14th, 2021 in Mumbai. Ankita Lokhande and her husband Vicky Jain won Season 1 of Smart Jodi on StarPlus in February 2022.
